I'm looking for the optimal way to create responsive landers today... Do people just use Dreamweaver? Certain scripts?
How can we be sure they work properly for all mobile devices?

I'd suggest you start with the following video made by caurmen where he makes a responsive lander in less than 10 minutes:

It takes a bit of HTML & CSS but it's definitely worth it.
