Large targeted Email list but need Help getting started
Last year I started a cosmetics line that was doing very well on facebook. In the process I collected 250k emails from optins and confirmed buyers. My list consists of women 25-65 who are interested in cosmetics. I know emailing has alot to do with your relationship with the customers. Im thinking about hitting the list with skin offers but obviously need to build some trust first. Was thinking about sending lots of free skin care content, videos, articles, ect for a while before I hit them with $ offers. Any thoughts on how I should approach this? Thanks

You pretty much got it right - heat up the list with valuable information first, then move on to selling. If you bomb the subscribers with offers straight away, you will sell some products, but the unsubscribe rate will be high and the response will be lower with every blast.
250k targeted emails with demo data sounds like a solid asset. I would personally start to develop a site too and use the email list to send them newsletter with new content. This way you could grow the list further and build a solid site - in case you have the resources to actually produce useful content that would be of interest to your list members.

Maybe use the list for FB LLA, this way you can make money instantly next to your e-mail funnel warm up process.

Provide your users with valuable content and blend in ads from time to time (native) instead of in the face ad pushing which will lead to high unsubscrube rates and falling response rates. Also suggest your users to bring other users and give them a reward if they do (unique discount codes, vouchers, etc) that way you can grow your list. Best is of course if you can link the database to a website so that users not only read your emails but also start visiting your (content) site or webshop.

Facebook - Look a Like Audience . If he/she uploads this list on FB, and create a similar target group with it, it's "easy" to get in conversions for low cost.

If you are looking to really build a relationship with your list and provide the most value while making revenue then the big thing to focus on is segmentation and data
You already mentioned your list is made up of opt-in and buyers. This is the first and easiest segment to create. In general a buyer list is worth multiple times more than a non-buyer list. So you should treat the two segments differently. If you decide to upload the list to FB for a LLA, your buyer segment "should" be able to find you more people like your buyers vs the rest of the list that are currently non-buyers.
When you are crafting your emails, you can tailor your message based on the fact that they have either bought something or not. If you are handy with code, you should be able to append the current LTV of each buyer as a data point to your list, so you can also segment and message based on the quality of buyer.
The quality of your segments also depends on how much data you have on each subscriber. You can buy it from a company like http://www.towerdata.com but you can get much better data by just asking people for the data. All it takes is a well designed quiz that actually appends data back to your ESP.
Remember that reason the FB is even able to create a LLA is because it has so many data points on each user.
With that covered you can begin to do data driven email marketing to your list that will product much better results than the usual untargeted blasts.
I usually tell people to build an auto responder sequence for hands free, but to also daily mailings (targeted hybrid blasts - educational stuff that sells) to the list. If you don't mail often enough, people start to forget who you are and why they are getting emails from you.
When you find an unusually good promo/blast you should slot it into your auto responder sequence for everyone who joined after the promotion. Or keep it in your swipe file for resending at a later date.
The thing to keep in mind is that even if you are getting 20-30% open rates, there are 70-80% of the people on your list that didn't open. So sending the exact same email again at a later date works unbelievably well. I will usually email the same email later on in the day (or next day) to unopens with a new headline or I'll swap it out. It mainly depends on how well the open and click rates were.
