When you look at your data and not happy with it, how soon do you make the adjustments? Do you wait till it reaches an X amount of clicks before changing things up? When is it too soon to make changes and too late?

You need to better define your question.
When to make optimisations and decisions depends entirely on the data you are looking at.
You should of course make decisions based on statistically significant differences where practical - you should check out the mobile cookbook as Caurmen goes over some of the typical maths/tools you would use for this.
Agreed, check the statistical significance guide by Caurmen and use one of the calculators he mentions.
Yessir, you don't want to just make mental judgements, follow a strict rule that you always use and Caurmens guide is fantastic on the subject.
